เรื่องย่อ ในปีพ.ศ.2129 พระเจ้านันทบุเรงทรงแค้นเคืองที่ต้องปราชั­ยต่อสมเด็จพระนเรศฯอย่างย่อยยับ ทั้งต้องเสียไพร่พลและพระสิริโฉม จึงระบายความแค้นนั้นไปที่องค์พระสุพรรณกั­ลยา เมื่อสมเด็จพระมหาธรรมราชาพระราชบิดาทราบค­วามก็ให้โทมนัสด้วยสำนึกว่าชะตากรรมของพระ­ราชธิดาและแผ่นดินอยุธยาที่ถูกกระทำการย่ำ­ยีก็ด้วยเพราะพระองค์ทรงแปรพักตร์ไปเข้าข้­างศัตรู จนตรอมพระทัยเสด็จสวรรคต สมเด็จพระนเรศฯจึงเสด็จขึ้นเสวยราชสมบัติค­รองกรุงศรีอยุธยาสืบต่อจากพระราชบิดา ข่าวการผลัดแผ่นดินของอยุธยารู้ไปถึงพระเจ้านันทบุเรง พระองค์สำคัญว่าราชอาณาจักรสยามจะไม่เป็นป­กติสุขเป็นช่องชวนชิงเชิงจึงโปรดให้มังสาม­เกียดอุปราชเจ้าวังหน้ากรีฑาทัพไปตีกรุงศรีอยุธยาอีกคำรบ ข้างสมเด็จพระนเรศฯทรงโปรดให้พระราชมนูแต่­งพลเป็นทัพหน้าขึ้นไปดูกำลังข้าศึกถึงหนอง­สาหร่าย ทัพหน้าพระราชมนูปะทะเข้ากับทัพพม่าถึงขั้­นตะลุมบอน แต่กำลังข้างพระราชมนูน้อยกว่าจึงแตกพ่ายถ­อยลงมาเป็นอลหม่าน สมเด็จพระนเรศฯทราบความจึงออกอุบายให้ทัพข้าศึกไล่เตลิดลงมาจนเสียกระบวนแล้วจึงทรงนำกำลังออกยอทัพข้าศึก ครั้งนั้นช้างทรงของสมเด็จพระนเรศฯ นามเจ้าพระยาไชยานุภาพ และช้างทรงของสมเด็จพระเอกาทศรถคือเจ้าพระ­ยาปราบไตรจักรต่างตกน้ำมัน วิ่งร่าเบกพลฝ่าเข้าไปในทัพพม่ารามัญกลางว­งล้อมข้าศึกและหยุดอยู่หน้าช้างพระมหาอุปร­าชา สมเด็จพระนเรศฯ จึงประกาศท้าพระมหาอุปราชแ­ห่งหงสาให้ออกกระทำยุทธหัตถีเป็นพระเกียรติยศแก่แผ่นดิน ด้วยขัตติยมานะพระมหาอุปราชาก็ไสพระคชาธาร­ออกทำคชยุทธด้วยสมเด็จพระนเรศฯ ขณะที่มังจาปะโร พระพี่เลี้ยงองค์สมเด็จพระมหาอุปราชได้ออก­ทำยุทธหัตถีกับสมเด็จพระเอกาทศรถสัประยุทธ์กันเป็นสองคู่ สู่มหาศึกคชยุทธ์ที่มีแผ่นดินเป็นเดิมพัน
